Determination of Km and Vmax for acetyl CoA, based on the generation of acetylated 1 MLAL-RRR 24 oligopeptide. Purified MBP-hNaa30p (80 nM) was incubated with 1MLAL-RRR24 oligopeptide at saturated levels and varying concentrations of acetyl CoA (4 – 40 μM) in acetylation buffer for 30 minutes at 37°C. A; Non linear regression analysis of the dose dependent curve generated based on the analysis of the acetylated oligopeptide absorption signal at 215 nm.
